    مستخلص: The insulin complement with gene therapy has been used as an experimental treatment for insulin dependent diabetes (IDDM). In the present study, we constructed naked plasmid DNA vector encoding recombinant human preproinsulin gene (pCMV-IN), and injected the plasmids (100 microg/mouse) intramuscularly combined with electroporation, to achieve the in vivo transfer of insulin gene in streptozotocin (STZ)-induced diabetic C57 mice. The expression of vector-derived insulin mRNA was detected with RT-PCR in transfected local skeletal muscles. The plasma insulin was elevated significantly in pCMV-IN injected diabetic C57 mice, which was complemented to the level similar to the intact normal control. The protein expression lasted for at least 35 days after the plasmid injection. Gene therapy with pCMV-IN plasmids considerably decreased the blood glucose level in STZ-induced diabetic mice from d 7 to d 35 by about 6 mmol/L. The gene therapy also reduced the mortality of severe diabetic mice significantly from 100% to 37% at the 6th week. Our results indicate that the direct intramuscular injection of naked plasmids encoding human preproinsulin gene achieves the effective expression of insulin. The restoration of insulin decreases blood glucose and increases the survival in severe diabetic mice. The gene therapy might be provided as a practical therapeutic approach to IDDM.

    مستخلص: Aim: To study the effects of indometacin (Ind) on joint damages.
    Methods: The volume of noninjected hind paw and interleukin-1 (IL-1) production from peritoneal macrophages and articular synoviocytes induced by lipopolysaccharides were assayed in adjuvant arthritis (AA) rats. Measurements of synovial fibroblast proliferative response and proteoglycan synthesis of cartilage from rabbits were used.
    Results: The secondary inflammatory reactions in AA rats on d 18, 21, and 24 were suppressed by i.g. Ind 2 mg.kg-1.d-1 for 9 d. Ind promoted IL-1 production from both macrophages and synoviocytes in AA rats. Ind 10 mumol.L-1 enhanced the proliferation of rabbit synovial fibroblasts and suppressed the proteoglycan synthesis of articular cartilage in response to IL-1 in vitro.
    Conclusion: Ind is unfavorable to the repair of joint destruction.

    مستخلص: Aim: To determine alterations of subtypes of myocardial adrenoceptors in senescence.
    Methods: Heart membrane preparations were made from 3- and 25-month old Wistar rats. Alpha 1- and beta-adrenoceptors were measured by radioligand, 125I-BE2254 and 125I-pindolol, binding assays, respectively.
    Results: In the old rat heart, alpha 1- and beta-adrenoceptor densities were declined from the young rats of 119 +/- 4 and 45.9 +/- 1.9 pmol L-1 to 70 +/- 6 and 36.4 +/- 1.6 pmol L-1 (P < 0.01), with a greater change in alpha 1-AR and in beta-AR. The ratio of alpha 1A/alpha 1B subtypes was decreased from the young rats of 39/61 to the old rats of 26/74 (P < 0.05).
    Conclusion: The cardiac adrenoceptors are decreased with different extents in the different subtypes in old rats.

    مستخلص: In the present study, we found that a novel alpha 1-adrenergic receptor subtype existed in rat heart by radioligand binding assay. This new alpha 1-adrenergic receptor subtype was different from alpha 1A-, alpha 1B-, alpha 1C-, and alpha 1D-subtypes reported recently. It had high affinity for WB4101, as the same of alpha 1A-subtype, but low affinity for 5-methyl-urapidil, and was insensitive to chlorethylclonidine.

    مستخلص: In the present study, we observed the effects of indomethacin, ibuprofen, naloxone and morphine on plasma CGRP concentration 3 h after endotoxin shock in conscious rats. The results showed that indomethacin, ibuprofen and naloxone significantly inhibited the elevation of plasma CGRP concentration as well as reduced the lesion in the intestine during endotoxin shock. However, morphine had no significant effect. The data are consistent with our hypothesis that prostaglandin, as one of the inflammatory mediators which increase in shock may be an important endogenous factor for triggering the CGRP release during endotoxin shock.

    مستخلص: [3H]Dihydroalprenolol ([3H]DHA) was used to label beta-adrenoceptors in intact lymphocytes isolated from rat spleen. The binding conditions were investigated by the methods of L9 (3(4)) orthogonal analysis, the results showed the optimum binding conditions were pH 7.7, 1 x 10(8) lymphocytes.ml-1 and 10 min at 25 degrees C for incubation. On these conditions the [3H]DHA binding to beta-adrenoceptors was rapid (T1/2 2 min) and readily reversed by propranolol 1 mmol.L-1 (T1/2 4 min). [3H]DHA saturation experiments indicated a single class of site with a KD of 7.2 +/- 2.2 nmol.L-1 and Bmax of 81 +/- 28 fmol/10(7) lymphocytes (n = 3). Computer analysis of competition experiments with isoproterenol and epinephrine revealed two classes of sites. One site had high affinity for the [3H]DHA and comprised 60 +/- 5% of the total number of sites, whereas the other site had a lower affinity. The affinity of the high affinity site was about 3 orders of magnitude higher than that of the lower affinity site.
