    مستخلص: Objective: Heterogeneity in circulating lipid concentrations in response to different dietary fat intakes may be due, in part, to gene polymorphism of cytokine related to inflammation. The objective was to determine whether dietary n-3 PUFAs intake influenced the effects of IL-6 -572 polymorphism on plasma lipids and apolipoproteins.
    Study Design: This cross-sectional study included 195 men and 386 women aged 40-65 y. DNA was isolated from white blood cell to determine the IL-6 -572 C-G polymorphisms by the polymerase chain reaction-logation detection reaction (PCR-LDR) method. Dietary fatty acids were assessed by measuring erythrocyte membrane fatty acids, and fasting plasma was collected to determine blood lipids and apoproteins.
    Results: The frequency of the G allele was 0.214. There were no significant differences in plasma lipids across genotypes (all p > 0.05). When men and women were grouped into tertiles by the erythrocyte membrane n-3 PUFAs composition; in the lowest group, men with G allele had 18% higher HDL-cholesterol concentrations than did C/C genotype (p < 0.05). Similar effects were observed for apo A. Adjusting for age, BMI and total PUFA did not alter this association. The interaction was not significant in women.
    Conclusion: Dietary n-3 PUFA intake influenced the effects of IL-6 -572 genotype on HDL-c concentrations in males.

    مستخلص: Objective: To investigate the effect of interleukin-6 (IL-6) and calcium sensing receptor (CASR) gene polymorphisms on bone mass accrual in Chinese adolescent girls.
    Methods: A total of 228 premenarche Chinese girls (9-11.5 years) were recruited for a 2-year study. Bone mineral densities (BMD) and Bone mineral contents (BMC) in the total body, total left hip including femoral neck, trochanter, intertrochanter and Ward's triangle area, and lumbar spine (L1-L4) were measured by DEXA. The -634C/G polymorphism of IL-6 gene was detected by PCR-restriction fragment length polymorphism and A986S polymorphism of CASR gene was detected by allele-special mutagenically separated amplication-PCR.
    Results: There were 176 available subjects when the 2-year study was completed. The -634C/G polymorphism of IL-6 gene and A986S polymorphism of CASR gene were significantly associated with bone mass accrual after adjusting the potential confounding factors. Girls with CC genotype of IL-6 -634G/C gene had higher percentage of increase in BMD of total body (P= 0.027) and femoral trochanter (P= 0.028) than those with CG+ GG genotypes. Girls with AA genotype of CASR gene had greater percentage of increase in L1-L4 lumbar spine BMC (P= 0.022) and Ward's triangle area BMD (P= 0.049) than their AS+ SS counterparts. Subjects with G allele of IL-6 gene in combination with S allele of CASR gene had less increase in BMC of femoral neck and L1-L4 lumbar spine.
    Conclusion: Adolescent girls carrying the G allele of IL-6 gene at -634C/G and S allele of CASR gene at A986S could potentially be the risk population of lower bone mass accrual.

    مستخلص: We assessed the main and interaction effects of interleukin-6 and estrogen receptor gene polymorphisms on bone mass accrual in Chinese adolescent girls. Bone mineral density (BMD) at the total body, lumbar spine (L1-L4), and total left hip were measured by dual-energy X-ray absorptiometry at baseline and follow-up. The -174G/C and -634C/G polymorphism of IL-6 gene, and PvuII and XbaI polymorphisms of the estrogen receptor (ER)-alpha gene, were determined. The -634C/G polymorphism of the IL-6 gene and PvuII polymorphism of ER-alpha gene were significantly associated with bone mass accrual after adjusting the potential confounding factors. Girls with pp genotype of ER-alpha gene had greater percentage accrual in BMD of total body (P = 0.010) and femoral intertrochanter (P = 0.038) than their PP and Pp counterparts. Girls with CC genotype of IL-6 -634G/C gene had higher percentage accrual in BMD of total body (P = 0.032) and femoral trochanter (P = 0.048) than their CG + GG counterparts. Significant interaction effects of IL-6 -634C/G polymorphism and ER-alpha PvuII polymorphism were observed on percentage change in BMD of total left hip (P = 0.009) and femoral intertrochanter (P = 0.007). The genotype CC (IL-6 -634C/G) x pp (ER-alpha PvuII) was associated with greater BMD accrual than other genotype combination in Chinese adolescent girls. We found that the IL-6 -634C/G and ER-alpha PvuII polymorphism were significantly associated with BMD accrual and that they have an interactional effect on BMD accrual in Chinese adolescent girls.

    مستخلص: Introduction and Hypothesis: A large number studies have examined the association between estrogen receptor alpha (ESR-alpha) gene polymorphisms and bone mineral density (BMD) in the Chinese population. We conducted a meta-analysis to assess their pooled effects.
    Methods: We searched for all published articles indexed in MEDLINE, the Chinese Biomedical Database, and the Chinese Journal Full-text Database from January 1994 to April 2006. Any cross-sectional study that tested the association between ESR-alpha PvuII or XbaI genotypes and BMD at the femoral neck or spine in Chinese women was included in the review. Data were extracted independently by two reviewers using a standardized data extraction form. Sixteen eligible studies involving 4,297 Chinese women were identified.
    Results: The overall frequencies of X and P alleles were 28% and 40%, respectively. The PvuII polymorphism was statistically significantly associated with BMD at the femoral neck (P = 0.038 for PP = Pp = pp) but not at the lumbar spine in all women. The BMD difference for the contrasts of PP versus Pp/pp genotypes was -0.0105 (95%CI, -0.0202 approximately -0.0008) g/cm(2) (P = 0.036). The XbaI polymorphism was not associated with BMD at the femoral neck or lumbar spine.
    Conclusion: The PvuII polymorphism had a very weak association with femoral neck BMD whereas XbaI polymorphism was unlikely to be a predictor of femoral neck or spine BMD in Chinese women.
