    الوصف: Ikigai, a Japanese term often referred to as “purpose in life” or “the feeling that life is worthwhile”, is a commonly used indicator of well-being in studies focused on the elderly population in Japan. It can be identified within the different realms like family, work, and friendship. However, as individuals age, their sense of ikigai is prone to diminish. Teachable agents (TAs) have been employed in educational settings for decades to facilitate learning by teaching students. Previous research has indicated that TAs can have a positive impact on self-esteem which is an important aspect of ikigai. TAs may hold the potential to address issues related to declining ikigai that are often associated with aging. By engaging in the act of teaching the agent, elderly individuals may experience a sense of being needed, consequently enhancing their self-esteem and potentially leading to an improvement in their ikigai levels. In this paper, we introduce a TA designed around the concept of ikigai, aiming at assisting the elderly in maintaining a high level of ikigai to support healthy aging. Based on a user study conducted using the phenomenographical approach, we demonstrate the effectiveness of our proposed TA design.

    الوصف: BackgroundSerous ovarian carcinoma (SOC) is considered the most lethal gynecological malignancy. The current lack of reliable prognostic biomarkers for SOC reduces the efficacy of predictive, preventive, and personalized medicine (PPPM/3PM) in patients with SOC, leading to unsatisfactory therapeutic outcomes. N6-methyladenosine (m6A) modification-associated long noncoding RNAs (lncRNAs) are effective predictors of SOC. In this study, an effective risk prediction model for SOC was constructed based on m6A modification-associated lncRNAs.MethodsTranscriptomic data and clinical information of patients with SOC were downloaded from The Cancer Genome Atlas. Candidate lncRNAs were identified using univariate and multivariate and least absolute shrinkage and selection operator-penalized Cox regression analyses. The molecular mechanisms of m6A effector-related lncRNAs were explored via Gene Ontology, pathway analysis, gene set enrichment analysis, and gene set variation analysis (GSVA). The extent of immune cell infiltration was assessed using various algorithms, including CIBERSORT, Microenvironment Cell Populations counter, xCell, European Prospective Investigation into Cancer and Nutrition, and GSVA. The calcPhenotype algorithm was used to predict responses to the drugs commonly used in ovarian carcinoma therapy. In vitro experiments, such as migration and invasion Transwell assays, wound healing assays, and dot blot assays, were conducted to elucidate the functional roles of candidate lncRNAs.ResultsSix m6A effector-related lncRNAs that were markedly associated with prognosis were used to establish an m6A effector-related lncRNA risk model (m6A-LRM) for SOC. Immune microenvironment analysis suggested that the high-risk group exhibited a proinflammatory state and displayed increased sensitivity to immunotherapy. A nomogram was constructed with the m6A effector-related lncRNAs to assess the prognostic value of the model. Sixteen drugs potentially targeting m6A effector-related lncRNAs were identified. Furthermore, we developed an online web application for clinicians and researchers (https://leley.shinyapps.io/OC_m6A_lnc/). Overexpression of the lncRNA RP11-508M8.1 promoted SOC cell migration and invasion. METTL3 is an upstream regulator of RP11-508M8.1. The preliminary regulatory axis METTL3/m6A/RP11-508M8.1/hsa-miR-1270/ARSD underlying SOC was identified via a combination of in vitro and bioinformatic analyses.ConclusionIn this study, we propose an innovative prognostic risk model and provide novel insights into the mechanism underlying the role of m6A-related lncRNAs in SOC. Incorporating the m6A-LRM into PPPM may help identify high-risk patients and personalize treatment as early as possible.

    الوصف: Abstract Background Few previous studies have introduced general techniques to overcome the “chopstick effect” in laparoendoscopic single-site surgery (LESS). We aim to investigate and highlight the key ergonomic methodologies for gynaecologic LESS based on the surgeon’s hands-on performance. Methods The first author surgeon A reviewed and analyzed the LESS procedures performed by herself and how she taught surgeon B from January 2021 to April 2022. The procedures were classified based on technical difficulty and learning periods, and the hands-on technical skills of LESS module were evaluated. Results Surgeon A conducted 580 LESS procedures, which were divided into the novice (n = 48) and intermediate (n = 33) periods, and the remaining cases were included in the routine period. We formed a special ergonomic LESS operating methodology: Maintain good LESS laparoscopic spatial sensation, keep hand-eye coordination, well cooperation between the main surgeon and the assistant; Experienced multiport laparoscopy surgery (MPS) skills, improve basic LESS technique: grasp, lift, transfer, place, blunt separating, coagulation and cutting. Coordination location, orientation, movements, and flexion or extension of shoulders, arms, elbow, wrist and finger joints; Maintain strength, tension and ambidexterity postures with joint and muscular efforts to control instruments. Surgeon B learned the above experiences by performing 39 LESS procedures under the guidance of surgeon A. Conclusion This educational research sheds light on the common challenges faced in LESS and presents the importance of ergonomic hands-on performance skills in improving surgical outcomes, which could serve as a guide for future training and education in LESS.

    الوصف: Predicting MYC and BCL2 double-expressor lymphoma prognosis using the system immune-inflammatory index (SII) and prognostic nutritional index (PNI) (DEL). From January 2015 to December 2021, 281 diffuse large B-cell lymphoma (DLBCL) wax blocks were used to make tissue chips. Screening double expressor lymphoma (DEL) instances involved immunocytochemistry and fluorescence in situ hybridization. Academic analysis used clinicopathological characteristics and follow-up data. SII, PNI, and DEL prognosis were correlated using univariate and multivariate cox regression analysis. The median age of 78 DEL patients is 60 (range: 43–74). SII and PNI cut-off values of 603.5, 3.07, and 144 predict PFS and OS well. Lower SII is associated with longer PFS (HR for SII = 0.34, 95% CI 0.15–0.76, P = 0.006; HR for NLR = 0.46, 95% CI 0.22–0.99, P = 0.048; HR for PLR = 0.39, 95% CI 0.17–0.94, P = 0.025; LMR = 0.39, 95%, CI 0.17–0.94, P = 0.025) and OS (HR for SII = 0.16, 95% CI 0.05–0.51, P = 0.005; HR for PNI = 0.20, 95% CI 0.06–0.62, P = 0.002). SII and PNI are promising predictors for twofold expressor DLBCL. Combining these increase prediction accuracy.

    الوصف: In microgravity, atoms can be prepared to a lower temperature and manipulated with a longer time. The Cold Atom Physics Research Rack (CAPR) in Chinese Space Station, which aims to prepare quantum degenerate gas for deep cooling and quantum simulation in microgravity, has been designed, realized, and validated. We built an integrated CAPR, consisting of a physical system, an all-fiber 780 nm laser system and a low phase noise 1064 nm fiber laser system, and an electrical driving and controlling system. With the CAPR, a 87Rb quantum degenerate gas with 1.2 × 105 atoms and a temperature below 30 nK was achieved on ground. In this article, we also show the robustness of the rack, based on the experimental results before and after the simulation of the vibration and impact during the launch.

    الوصف: 5-methylcytosine (m5C) post-transcriptional modifications affect the maturation, stability, and translation of the mRNA molecule. These modifications play an important role in many physiological and pathological processes, including stress response, tumorigenesis, tumor cell migration, embryogenesis, and viral replication. Recently, there has been a better understanding of the biological implications of m5C modification owing to the rapid development and optimization of detection technologies, including liquid chromatography-tandem mass spectrometry (LC-MS/MS) and RNA-BisSeq. Further, predictive models (such as PEA-m5C, m5C-PseDNC, and DeepMRMP) for the identification of potential m5C modification sites have also emerged. In this review, we summarize the current experimental detection methods and predictive models for mRNA m5C modifications, focusing on their advantages and limitations. We systematically surveyed the latest research on the effectors related to mRNA m5C modifications and their biological functions in multiple species. Finally, we discuss the physiological effects and pathological significance of m5C modifications in multiple diseases, as well as their therapeutic potential, thereby providing new perspectives for disease treatment and prognosis.

    الوصف: Plain English summary The Coronavirus Disease 2019 (COVID-19) is a global pandemic and posed serious challenges in many countries. This study aimed to investigate the psychological distress level of the primary caregivers of eating disorder (ED) offspring during the COVID-19 pandemic, and to explore potential influencing factors. From March 6 to April 20, 2020, 378 questionnaires for primary caregivers of ED offspring and 1163 questionnaires for primary caregivers of healthy offspring were collected through an online crowdsourcing platform in mainland China. All primary caregivers were evaluated for depression, anxiety, perceived stress and social support. 315 primary caregivers of ED offspring and 315 matched primary caregivers of healthy offspring were included in the statistical analysis. We compared the primary caregivers of ED offspring with the primary caregivers of healthy offspring for depression, anxiety, perceived stress and social support. We found that primary caregivers of ED offspring experienced more psychological distress than that of primary caregivers of healthy offspring. ED caregivers with high perceived stress may have higher levels of depression and anxiety. ED caregivers with high social support, no mental illness and no family conflicts may have lower levels of depression. ED caregivers with high social support, no family conflicts, and high years of education may have lower levels of anxiety.

    الوصف: As the real load is very complicated during the process of harvester operation,it is of great significance for the prediction of fatigue life to describe the load characteristics and distribution form accurately. In order to obtain the probability density distribution model of the load cycles. Take RF-half shaft torque as the research object,the mixture Gaussian fitting method based on EM was suggested. n Code software was used for rain flow counting and the load cycle was described with the vector S.The cluster number and cluster center loading were got from the clustering of the loading cycle by the SPSS software,and using EM algorithm to resolve the mean and covariance matrix of Gaussian component. The index value of SSE and RSME are close to0,but R and Raare both up to 0. 85 based on the index model. It showed that the Gaussian mixture indeed fitting out of the original data of load-cycle. The results of research can provide a theory evidence for the fatigue life analysis and load spectrum extrapolation.
