Abstract (English): The aim of the study was to investigate the effect of n-butyl baicalin on H2O2-induced oxidative stress in IPEC-J2 cells and its molecular mechanism. The suitable concentration of n-butyl baicalin was determined by CCK-8 assay. The levels of reactive oxygen species (ROS), the activity of superoxide dismutase (SOD) and catalase (CAT), and the content of malondialdehyde (MDA) were detected by ELISA, and the mRNA expressions of HO-1 and NQO-1 were detected by qPCR. The expression levels of Nrf2 protein in cytoplasm, Nrf2 protein in nucleus and HO-1 were detected by WB method, and the entry of Nrf2 protein into nucleus was detected by immunofluorescence method. The results showed that the suitable concentration of BE was 10, 20, 40 mmol/L. Compared with the control group, the content of ROS and MDA in H2O2 group were significantly increased ( P<0.05), the activity of SOD and CAT were significantly decreased ( P<0.05), the cytoplasmic Nrf2 protein and nucleus Nrf2 protein were significantly increased ( P<0.05), and the expression of HO-1 mRNA was significantly increased ( P<0.05). The expression level of NQO-1 mRNA was significantly decreased ( P<0.05). Compared with H2O2 group, medium and high doses of BE significantly decreased ROS and MDA content ( P<0.05). Low, medium and high doses of BE significantly increased SOD and CAT activity ( P<0.05). Medium dose of BE significantly increased the expression of Nrf2 protein ( P<0.05). Low, medium and high doses of BE significantly increased the expression of Nrf2 protein in nucleus ( P<0.05). Medium and high doses of BE significantly increased the expression level of HO-1 mRNA ( P<0.05), and low, medium and high doses of baicalin significantly increased the expression level of HO-1 protein. Low, medium and high doses of BE significantly increased the expression level of NQO-1 mRNA ( P<0.05). The study indicates that BE played an antioxidant role by activating Nrf2 signaling pathway. Abstract (Chinese): 试验旨在探讨黄芩苷酯化物 (BE) 对 H2O2 诱导的 IPEC-J2 细胞氧化应激的缓解作用及其分子机制。 采用 CCK-8 法检测细胞活性筛选出黄芩苷酯化物的适用浓度; 采用 ELISA 法检测细胞中活性氧 (ROS) 水平、 超氧化物歧化酶 (SOD) 活性、过氧化氢酶 (CAT) 活性、丙二醛 (MDA) 含量; 采用 qPCR 法检测细胞中HO-1 和 NQO-1 mRNA 的表达量; 采用 WB 法检测胞浆 Nrf2 蛋白、胞核 Nrf2 蛋白和 HO-1 的蛋白表达水平; 免 疫荧光法检测 Nrf2 蛋白入核情况。结果显示, BE 的适用浓度为 10、20、40 mmol/L; 与对照组相比, H2O2 组中 ROS、MDA 含量显著升高 ( P<0.05) ; SOD、CAT 活性显著下降 ( P<0.05) ; 胞浆 Nrf2 蛋白与胞核 Nrf2 蛋白表达 量显著升高 ( P<0.05) ; HO-1 mRNA 的表达量显著升高 ( P<0.05) ; NQO-1 mRNA ( P<0.05) 表达量显著降 低。与 H2O2 组相比, 中、高剂量 BE 显著降低 ROS、MDA 含量 ( P<0.05) ; 低、中、高剂量 BE 显著提高 SOD、 CAT 活性 ( P<0.05) ; 中剂量 BE 显著升高胞浆 Nrf2 蛋白的表达 ( P<0.05) ; 低、中、高剂量 BE 显著升高胞核 Nrf2 蛋白的表达 ( P<0.05) ; 中、高剂量 BE 显著升高 HO-1 mRNA 的表达量 ( P<0.05), 低、中、高剂量均可显著升 高 HO-1 蛋白表达; 低、中、高剂量 BE 显著升高 NQO-1 mRNA 表达量 ( P<0.05) 。研究表明, BE 可以通过激 活 Nrf2 信号通路发挥抗氧化作用。 [ABSTRACT FROM AUTHOR]
