مستخلص: Butyrivibrio fibrisolvens bacteria of rumen origin were isolated to study their conjugated linoleic acid (CLA) production potential with the supplementation of pure linoleic acid (LA) in selective media (ATCC broth). Butyrivibrio cultures were isolated from the rumen liquor of fistulated buffalo and are confirmed by different biochemical tests and PCR using a genus-specific primer. Three strains namely 49, 0/10, MZ3 of B. fibrisovens showed the positive signal in the form of discrete and distinct 220, 220 and 235 bp bands, respectively. Isolated bacteria were used in in vitro experiments with pure LA at different concentrations (50, 100, 150, 200 and 250 µg/ml of media) and incubation period (0, 8, 12, 18 and 24 hrs) to study CLA production potential of the bacteria. The highest CLA production was recorded at 18 h of incubation period with the concentration of 200 µg of LA (65.52 µg/ml). However, it decreased (42.24 µg/ml) at 250 µg of LA may be due to the toxic effect of LA on microbial growth. [ABSTRACT FROM AUTHOR]
