Abstract (English): Objective: A high performance liquid chromatographytandem mass spectrometry (HPLC-MS/MS) method was established for rapid determination of empagliflozin, [magliptin, saxa-gliptin and alogliptin in health foods. Methods: Target compounds in samples were ultrasonically extracted by methanoU separated by reverse liquid chromatography and detected by electrospray ionization (ESI) using multiple reaction monitoring (MRM) mode. The quantitation was carried out by external standard method of matrix calibration curve. Results: Good linear relationship displayed in the concentration range of 2〜100 yg/L for sax-agliptin and alogliptin, with the limits of quantitation (LOQs) of saxagliptin and alogliptin of 0.10 mg/kg. At three spiked concentration levels of 0.1, 0. 2 and 1. 0 mg/kg, the recoveries were 79.2%〜113. 0% $ with relative standard deviations (RSDs) of 0.9%〜8.4%. The linear ranges of empagliflozin and linagliptin were 20〜500 yg/L with LOQs of empagliflozin and linagliptin were 1.00 mg/kg. The recoveries were 77.5 % 〜111.1 % with relative standard deviations(RSDs) of 2.5%〜9.4% at three spiked concentration levels of 1.0, 2.0 and 10.0 mg/kg. Conclusion: The method is simple, rapid, accurate and suitable for identification and determination of 4 anti-diabetic medicines in health foods. [ABSTRACT FROM AUTHOR]
Abstract (Chinese): 目的:建立高效液相色谱 串联质谱法快速测定保 健食品中4种降血糖类药物(恩格列净、利格列汀、沙格 列汀、阿格列汀)的方法。方法:样品经甲醇超声提取, 用 反相高效液相色谱分离, 在多反应监测扌莫式下测定, 外标 法定量。结果:沙格列汀和阿格列汀在2-100 pg/L内 线性关系良好, 定量限为o. 10 mg/kg;在0. 1, 0. 2, 1.0 mg/kg加标水平下, 回收率为79.2%〜113.0%,相对 标准偏差为0.9%〜&4%;恩格列净和利格列汀在20〜 500 pg/L内线性关系良好, 定量限为1.00 mg/kg;在1.0, 2.0,10.0 mg/kg加标水平下, 回收率为77.5%〜111.1%, 相对标准偏差为2.5%〜9.4%。结论:该方法简便、快速、 准确, 可用于保健食品中4种降血糖类药物的确证与定 量测定.
