مستخلص: The ethnobotanical survey revealed that Ampelocisuss indica (AI) has many reported traditional uses such as jaundice, pustules, carbuncles, blood purifier, diuretics, etc. The main purpose of this research is to evaluate the hepatoprotective activity of methanolic extract of Ampelocisuss indica (MEAI) in CCl4 induced hepatotoxicity in rats. The protective effect of MEAI was evaluated by measuring biochemical parameters such as ALP, SGOT, SGPT, and total bilirubin. Total phenolic content, in-vitro antioxidant activity and GC-MS analysis of MEAI were conducted. Further antioxidant activity of MEAI was determined by lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) levels in the supernatant of liver homogenate. MEAI at doses, 250mg/kg and 500mg/kg produced significant (P<0.05) hepatoprotection by decreasing the levels of serum enzymes, bilirubin, and lipid peroxidation, while they significantly increased the levels of glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) in a dose-dependent manner. The hepatoprotective and antioxidant activity of MEAI was comparable to that of the standard drug Silymarin. Histopathological studies showed good recovery in a dose-dependent manner. GC-MS analysis showed that MEAI contains 10 different types of volatile constituents and among these Hexadecanoic acid ethyl ester is a major constituent. Antioxidant and anti-inflammatory activity of Hexadecanoic acid ethyl ester has been reported. Hence the hepatoprotective activity of MEAI may be due to the presence of Hexadecanoic acid ethyl ester.
