Abstract (English): The experiment was to screen strains to produce protease by fermenting silk reeling wastewater. The strain Z416 was screened by sampling from natural environment. The strain was identified as Paenarthrobacter nitroguajacolicus by morphological observation and molecular biology. The experiment further optimized the combination of enzyme production conditions of strain through single factor and orthogonal design experiments. The results showed that the optimal fermentation scheme was lactose addition amount of 4% in fermentation broth, initial pH value of fermentation medium of 7.5, inoculum amount of 16%, the speed of the shaking table of 180 r/min, fermentation temperature of 33 ℃ and fermentation time of 84 h. Under these conditions, the enzyme activity of strain Z416 reached 17.89 U/mL, which was 5.57 times that of 3.21 U/mL before optimization. The study indicates the protease has the potential to be used in poultry and pig feed to improve the growth performance of livestock and poultry. [ABSTRACT FROM AUTHOR]
Abstract (Chinese): 试验旨在研究筛选菌株发酵缫丝废水生产蛋白酶。试验从自然环境取样, 筛选得到能利用缫丝废水 发酵产蛋白酶的菌株Z416, 形态学观察和分子生物学鉴定该菌株为硝基愈创木胶类节杆菌(Paenarthrobacter nitroguajacolicus), 并通过单因素和正交设计试验优化了该菌株的产酶条件组合。结果显示, 发酵液乳糖添加量 4%、发酵培养基初始pH 值7.5、接种量16%、摇床转速180 r/min、发酵温度33 ℃、发酵时间84 h 为最优发酵方 案。在此条件下, 菌株Z416 酶活力达到17.89 U/mL, 是优化前酶活(3.21 U/mL) 的5.57 倍。研究表明, 该蛋白 酶具有应用到禽和猪饲料中的潜力, 可以改善畜禽的生长性能.
