مستخلص: The present experiment, "Studies on evaluation of Inorganic Chemosterilants in adult male rats" was undertaken in a total of 24 male albino Wistar rats, which were randomly divided into control group (I) and three (3) treatment groups (II, III and IV) consisting of six (6) animals in each group for a period of 30 days. The rats in the groups I, II, III and IV were given single intratesticular injection of viz., normal saline, 20% Sodium Chloride (NaCl), Calcium Chloride (CaCl2) and Zinc gluconate solutions respectively for chemical sterilization. The mean body weight (g) values of rats in group II, III and IV on 0, 5th, 10th, 15th, 20th, 25th and 30th day did not vary significantly (P>0.05) when compared to group I. The mean values of hematologic parameters viz., the mean hemoglobin concentration level-g%, PCV-% values, TEC (millions/ìL) values and TLC (thousands/mL) values and the mean values of the biochemical parameters viz., Serum Alanine Transaminase (ALT-IU/L), Serum Aspartate Transaminase (AST-IU/L), Alkaline Phosphatase (ALP-IU/L), Serum Total Proteins (TP-g/dL), Serum Albumin (ALB-g/dL), Blood Urea Nitrogen (BUN-mg/dL) and Creatinine (mg/dL) values in group II, III and IV on 30th day of post injection did not differ significantly (P>0.05) when compared with the group I. A significant (P<0.05) decrease in the mean values of Testosterone (ng/mL) hormone in group III and IV rats on 15th and 30th day of post injection were recorded when compared to group I and II rats. The mean values of Testicular weight (mg), Epididymis weight (mg), Seminal vesicle weight (mg), Prostate weight (mg), Length of testis (mm), Width of testis (mm), Sperm motility (%) and Sperm count (millions/mL) in group III and IV rats on 30th day of post injection was showed significant (P<0.05) decrease when compared to group I and II rats. Haematoxylin and Eosin stained testes tissue sections of group I rats as control on 30th day post injection revealed normal testicular structure where as in group II rats resulted in severe degenerative changes in testicular seminiferous tubules, degeneration of testicular microstructure with coagulative necrosis and all testicular tissue underwent severe destruction with massive infiltration of immune cells leucocytes whereas as in group III rats resulted complete derangement of tubular architecture without any distinct boundary between the tubular and extratubular compartment along with infiltration of a large number of leucocytes throughout the testicular tissue and heavy mononuclear cellular infiltrations were seen in the interstitial tissue. Notable appearance of fibrous tissue was also observed throughout the testicular sections and in group IV rats resulted in high frequency of syncytial giant cells, inflammation, germ cell degeneration and necrosis, increased intertubular tissue (IT), thickening of the tunica albuginea, dystrophic calcification, congestion, tubular degeneration with vacuolated sertoli cells, and intertubular tissue (IT) showing macrophages and fibroblast proliferation. Some areas in the intertubular tissue (IT) showed necrotic Leydig cells. It was concluded that intratesticular injection of calcium chloride (CaCl2) and zinc gluconate might serve as a way of chemosterilization and may be considered as an alternative to surgical castration in male animals.
