مستخلص: Maerua oblongifolia(Forssk.) A. Rich. is a valuable medicinal plant used in many ayurvedic formulations, it has become threatened in Telangana State, India, due to the overexploitation by local tribal people and drug manufacturers. Since this species has not been extensively studied at the molecular level, an effort has been made in the current investigation to evaluate genetic diversity through RAPD markers. RAPD profiling generated 65 amplified bands from 12 decamer primers, of which 14 (21.53%) loci were polymorphic. The total amplification products per primer ranged from 4 (OPM-1) to 8 (OPM-7), with an average of 5.41 bands per primer. PIC values ranged from 0.06 (OPM-6 and OPM-9) to 0.23 (OPM-1). The amplified products of the 12 decamer primers collectively showed a similarity matrix index between the six accessions of M. oblongifoliaranging from 0.83 to 0.96. All the accessions were divided into two clusters by UPGMA cluster analysis based on the Jaccard similarity coefficient. Cluster-I comprises a single accession, while cluster-II comprises five accessions in two sub-clusters. Based on the detailed molecular characterization of M. oblongifolia, it can be assumed that understanding the genetic diversity among accessions will be critical for plant conservation and restoration. Furthermore, the molecular markers identified in this investigation will aid in the authenticity of this species to prevent adulteration in herbal medicine.
