الوصف: Using optical spectroscopy, we have studied the models of lipid peroxidation in mitochondrial membranes isolated from the hearts of rats. The oxidative efficiency of tested iron-containing complexes was assessed by the level of TBA-reactive compounds formed in the samples as the main secondary products of peroxidation. EPR spectroscopy of spin labels was used to study the changes in structural and conformational characteristics of mitochondrial membrane lipids in the presence of oxidation initiators. Mitochondrial suspensions were incubated with prooxidants, mostly myoglobin and cumene hydroperoxide, to detect the changes. In this study, we used spin labeled molecules of 5-doxylstearic and 16-doxylstearic acid with paramagnetic NO-fragments attached to different sites of the hydrocarbon chain. With the use of oxidation models, it was possible to obtain preparations of mitochondrial membranes with different degrees of oxidative modification. It was found that the combination of the initiators of oxidation, i.e., cumene hydroperoxide (or tert-butyl hydroperoxide), resulted in the failure of paramagnetiс centers of the spin-labeled membrane.
