Itsnatural polymorphism has not been studied atthe quasispecies level. In the present work, thegenetic heterogeneity of the NS3 protease genewasanalyzedin17HCVgenotype1(5subtypes1aand 12 subtypes 1b) samples collected frominfectedpatientsbeforeanti-viraltherapy.Atotalof 294 clones were sequenced. Although theprotease NS3 is considered to be one of the lessvariable genes in the HCV genome, variability ofboth nucleotide and amino acid sequences wasfound. In variants belonging to 1a and 1b sub-types, 224 and 267 of 543 positions showed oneor more nucleotide substitutions, respectively.Fortyand 74of the181NS3aminoacid positionsshowedatleastonemutationinHCV-1aandHCV-1bisolates,respectively.Mostsubstitutionswereconservative. This substantial polymorphism ofthe NS3 protease produced by HCV-1a and HCV-1b suggests that, despite the numerous func-tional and structural constraints, the enzymeis sufficiently flexible to tolerate substitutions.J.
